Abstract Introduction The long saphenous vein (LSV) is frequently used in cardiac surgery; however, its use complicated by late stenosis or occlusion due to the development of intimal hyperplasia (IH). TGF-β has been implicated process IH however impact acute haemodynamic changes on activation endothelial-to mesenchymal transition (EndMT) not assessed and it's focus this study. Purpose To assess role haemodynamics veins implanted into arterial circulation EndMT. Methods Surplus LSV were exposed using a perfusion bioreactor. Changes EndMT markers at RNA protein level evaluated quantitative real time PCR, RNAScope immunofluorescence. Results exposure ex-vivo induced significant increase inflammatory marker IL-8 expression transcription factor TWIST1 (both p≤0.01, Figure 1A) endothelium. Furthermore, immunostaining demonstrated pSMAD (p≤0.01, 1B) acutely endothelium after 45 minutes haemodynamics.
